Urea Broth Base
Urea Broth Base is a liquid medium used for differentiating Enterobacteriaceae, especially Proteus, based on urease activity. This medium, buffered with Di- and Monopotassium Phosphates, contains Urea and Phenol Red as pH indicator. Urease activity alkalinizes the environment, thus producing a color shift from pale yellow to pink-red. Yeast Extract provides the nitrogenous sources essential for bacterial growth. There is a choice of 3 sizes - 500g, 2kg, and 10kg.
PREPARATION: Mix 39.6 grams of the medium in one liter of purified water until evenly dispersed. Heat with repeated stirring to dissolve completely. Sterilize by filtration. DO NOT BOIL OR AUTOCLAVE.
